The officials say the Wednesday strike hit a vehicle and also wounded four people in the Kani Guram section of South Waziristan.
The US has escalated its missile campaign since August despite Pakistani protests.
South Waziristan is the main base for Baitullah Mehsud, the top Pakistani Taliban commander.
The two officials spoke on condition of anonymity because they are not authorized to speak to media.
